چکیده
Long non?coding RNAs (lncRNAs) are involved in the occurrence and progression of numerous types cancer. The aim present study was to evaluate effect lncRNA maternally expressed gene 3 (MEG3) on migration invasion non?small cell lung cancer (NSCLC) H1299 PC9 cells. Reverse transcription?quantitative (RT?q)PCR analysis showed that MEG3 downregulated NSCLC Additionally, bioinformatics indicated sponges microRNA (miR)?21?5p; miR?21?5p predicted target phosphatase tensin homolog (PTEN) 3'?untranslated region sequence. overexpression led suppression PTEN upregulation cells, as detected by RT?qPCR. Subsequently, western blot confirmed enhanced expression levels inhibited PI3K/AKT signaling pathway These effects were attenuated miR?21?5p. Dual luciferase assay supported sponging validated direct interaction between PTEN. Furthermore, Transwell demonstrated had an inhibitory invasion. also mediated epithelial?to?mesenchymal transition significantly enhancing E?cadherin decreasing N?cadherin, Vimentin matrix metalloprotein 9 analysis. changes partially reversed mimic. results acted a tumor suppressor via miR?21?5p, which, turn, PTEN, part pathway. have suggested potential novel therapeutic for treatment.

Long non-coding RNA FOXO1 inhibits lung cancer cell growth through down-regulating PI3K/AKT signaling pathway
Objective(s): Lung cancer is one of the most common malignant tumors, which seriously threatens the health and life of the people. Recently, a novel long non-coding RNA (lncRNA) termed lncFOXO1 was found and investigated in breast cancer. However, the effect of lncFOXO1 on lung cancer is still ambiguous. The current study aimed to uncover the functions of lncFOXO1 in l...
متن کاملLong noncoding RNA MEG3 suppressed endothelial cell proliferation and migration through regulating miR-21.
Long non-coding RNAs (lncRNAs) act critical roles in many biological processes, including cell proliferation, apoptosis, development, invasion and migration. LncRNA maternally expressed gene 3 (MEG3) is found to be downregulated in several tumors; however, its role in the atherosclerosis is still unknown. In the present study, we demonstrated that MEG3 expression level was downregulated in the ...
متن کاملshRNA-mediated downregulation of α-N-Acetylgalactosaminidase inhibits migration and invasion of cancer cell lines
Objective(s): Extracellular matrix (ECM) is composed of many kinds of glycoproteins containing glycosaminoglycans (GAGs) moiety. The research was conducted based on the N-Acetylgalactosamine (GalNAc) degradation of ECM components by α-N-acetylgalactosaminidase (Nagalase) which facilitates migration and invasion of cancer cells. This study aims to investigate the effects of Naga-shRNA downregula...
متن کاملLong Noncoding RNA H19 Inhibits Cell Viability, Migration, and Invasion Via Downregulation of IRS-1 in Thyroid Cancer Cells
Thyroid cancer is a common endocrine gland malignancy which exhibited rapid increased incidence worldwide in recent decades. This study was aimed to investigate the role of long noncoding RNA H19 in thyroid cancer. Long noncoding RNA H19 was overexpressed or knockdown in thyroid cancer cells SW579 and TPC-1, and the expression of long noncoding RNA H19 was detected by real-time polymerase chain...
متن کاملThe histone demethylase JMJD1A induces cell migration and invasion by up-regulating the expression of the long noncoding RNA MALAT1
Patients with neuroblastoma due to N-Myc oncogene amplification have a high frequency of tumor metastasis. However, it is not clear how N-Myc induces cell migration, invasion and metastasis. The histone demethylase JMJD1A activates gene transcription by demethylating the lysine 9 residue of histone H3 (H3K9) at target gene promoters. The long noncoding RNA MALAT1 induces lung cancer cell migrat...
